چکیده

A rapid seminested polymerase chain reaction (PCR) method for the specific, sensitive detection of virulent Shigella spp. in spiked environmental water samples was developed. A set of primers specific for the invasion plasmid antigen gene (ipaH) of virulent Shigella spp. and enteroinvasive Escherichia coli produced a 620-bp fragment that was used as template for the seminested primer pair delineating a 401-bp fragment. By using agarose gel electrophoresis for detection of the seminested PCR-amplified products, a detection limit of 1.6 10 cfu S. flexneri was obtained with amplification reactions from crude bacterial lysates. The PCR procedure coupled with an enrichment culture incubated for 6 h detected as few as 1.6 S. flexneri organisms in pure culture. Treated sewage, ground, surface and drinking water samples collected from various sources were seeded with S. flexneri and incubated in GN broth for 6 h before detection by seminested PCR. A detection limit lower than 14 cfu/ml was achieved in some water samples. The results indicate that the described seminested PCR has the advantage of a rapid turnaround time and it fulfills the requirements of sensitivity and specificity for use in an environmental laboratory. # 2001 Elsevier Science Ltd. Breaking down the time limit constraints for conventional compression–permeation (C–P) cell test, this work has, for the first time, experimentally evaluated the cake characteristics of viable wasteactivated sludge subject to polyelectrolyte flocculation and to freeze/thaw treatment under a pressure range of 25–200 kPa. There exists a threshold pressure exceeding which the cake structure would ...

This work experimentally elucidates the effects of ultrasonic treatment on the physical, chemical, and biological characteristics of a waste-activated sludge. A critical ultrasonic power level exists above which, accompanied with the release of divalent cations from the sludge body, the floc structure effectively disintegrated, microbial level acceptably disinfected, and particulate organic com...

In order to elucidate and quantify nitrogen transformations occurring during aerobic treatment of pig slurry, two laboratory experiments were carried out with contrasting levels of aeration, high level (experiment 1) and low level (experiment 2) of aeration. During these experiments, after reaching steadystate conditions, a single pulse of NO3 -N tracer was added directly to the reactor. When n...

The rates of adsorption of three commercial reactive dyes and Cu(II) from water in the absence and presence of complexing agents using chitosan were measured at 308C. Three simplified kinetic models, i.e., pseudo-first-order, pseudo-second-order, and intraparticle diffusion, were tested to investigate the adsorption mechanisms. It was shown that the adsorption of reactive dyes and Cu(II) in the...

Six upflow anaerobic sludge blanket (UASB) reactors were concurrently operated for 146 d to examine the effects of calcium on the sludge granulation process during start-up. Introduction of Ca at concentrations from 150 to 300mg/l enhanced the biomass accumulation and granulation process. The calcium concentration in the granules was nearly proportional to the calcium concentration in the feed,...
